Industrial fermentation involves a sample a tank of controlled pH, aeration, agitation, temperature. It requires more preliminary steps than other bioprocess workflows. Common microorganisms used in this process are Chinese hamster ovary cells (CHO), yeast, fungi, and bacteria. Masterflex® peristaltic pumps are useful in this workflow and are recommended over diaphragm pumps. Peristaltic pumps provide accurate dosing capabilities and, unlike diaphragm pumps, have an isolated and unconvoluted fluid path. This makes maintenance easy and ensures no product gets stuck in a crevice.
Typical harvest methods include Tangential Flow Filtration (TFF) or centrifugation as a means of concentrating the target compound. In either of these methods, quality and purity are paramount. Wetted materials (any material touching the fluid) must be validated USP Class VI and sterilized. Peristaltic pumps remain accurate at higher pressures, making them ideal for filtration centric workflows.
Affinity and ion-exchange chromatography are used to remove metals and reaction by-products along with additional methods of filtration. Recently, single-use tube component assemblies have become more prevalent in this workflow as higher throughputs and versatility on the process floor are demanded. The isolated fluid flow path of peristaltic pumps, such as Masterflex® pumps, is critical to this workflow. These pumps mitigate the potential for contamination and reduce the required material validation.
This critical process — highly regulated with global regulatory and compliance requirements — transfers the sterile product, now in its most valuable form, to a sterile vial or bottle before it is packaged and distributed or sent to lyophilization (freeze-drying) before packaging. Peristaltic pumps are the top choice for their ability to provide easy change-overs, reduce cross-contamination, and gently pump shear-sensitive fluids, large molecules, and live cells. The valuable end product is contained in the tubing, never touching any part of the pump.
